SUMMARY
In this study, Agrobacterium-mediated transformation protocol via young corn embryo was proposed and optimized. A. tumefaciens bacterial density appropriate values is OD = 0.8, the time optimal infection is 30 minutes, the acetosyringone concentration is 150 μM, the age of the embryo is 10-12 days old (size 0.9- 1.3 mm) optimized for capable of receiving gene and rebirth. The threshold for selective transgenic embryo is 50mg / ml kanamycin antibiotic. The successfully results of regeneration and transferred gus genes in to LVN99 maize is the basis for the study of transgenic defensin gene in to maize in Vietnam served generate transgenic maize lines that are resistant to weevils.
Keywords: A. tumefaciens, gus gene, regeneration, young corn embryo, Zea mays.
