High performance liquid chromatography with diode array detector (HPLC-DAD) was developed for the quantitative determination of O-methyl furodysinin lactone (1) in methanol extract of the sponge Dysidea fragilis (Montagu, 1818). The separations were effected by using a mobile phase of acetonitrile and water. The gradient elution of the mobile phase was 5-20% ACN in 0-12 min, 20-60% ACN in 12-50 min, 60-75% ACN in 50-65 min, 75-95% ACN in 65-95 min. The flow rate was 0.5 ml/min and the detector wavelength was set at 210 nm. This method was validated in terms of compatibility, specificity, linearity, precision, accuracy, limit of detection and limit of quantitation.
