ffects. Indole alkaloids can exert anti-hepato-
carcinogenesis e
ffects by affecting cell apoptosis, proliferation, cell
cycle metastasis and angiogenesis. Among them, promoting apoptosis
of tumor cells is most obvious. The role in
fighting against liver cancer
is mainly achieved by regulating the pathways associated with tumor
development, such as the NOD1 signal pathway, AKT pathway anad
WOWX-dependent pathway (
Fig. 5
).
2.4. Terpenoids alkaloids
Paclitaxel, a naturally occurring diterpenoid derived from the bark
and trunk of Yew Paci
fic, contributes to inducing and promoting tu-
bulin assembly and polymerization. Furthermore, it prevents depoly-
merization, increases the stability of tubulin,
finally helping to inhibit
the growth of tumor cells [
125
,

]. Paclitaxel has been used to treat a
variety of cancers, including breast cancer, ovarian cancer, lung cancer,
bladder cancer, head cancer, sarcoma and liver cancer [
127
]. Paclitaxel
markedly inhibits not only the proliferation of HCC cells, but also the
viability of HCC cells such as HLE, L-02 and Bel 7402 cells in a time-
and dose-dependent manner [
114
]. Alpha-fetoprotein (AFP), re-
cognized as an early biomarker, has been used for a long time in the
diagnosis of HCC and in follow-up systemic and locoregional therapies
[

]. Besides, the apoptosome number is increased in Bel 7402 cells
following treatment with paclitaxel. However, if transfected with AFP-
siRNA vectors after treatment with paclitaxel, the number of apopto-
some would increase more obviously [
114
]. Paclitaxel promotes the
cleavage of Caspase-3 and inhibits the expression of Ras and survivin in
HLE and L-02 cells. Silenced expression of AFP signi
ficantly facilitates
cleavage of Caspase-3 and suppresses the expression of Ras and survivin
in Bel 7402 cells [

]. Apart from this, the anti-AFP single-chain
variable fragment (scFv) induces growth inhibition of AFP-expressing
HCC cell lines by induction of apoptosis and cell cycle arrest [

].
These results indicate that paclitaxel can promote liver cancer cell
apoptosis and cell cycle arrest, and target AFP by inhibiting its ex-
pression or speci
fic antibodies, therefore accelerating this effect.
However, as we all know, paclitaxel is a water-insoluble drug with in-
solubility of approximately 0.006 mg/mL, thus generating low bioa-
vailability. Nonetheless, paclitaxel also exerts excellent anti-cancer ef-
fect, which could be used as the
first-line drug via combination with
other drugs or by means of some new formulation technique. Some
researchers have constructed an e
ffective nanoparticle system for car-
rying paclitaxel and IR780 in order to combine PTT/PDT (photothermal
therapy/photodynamic therapy) and chemotherapy to treat liver cancer
[

]. Paclitaxel and IR780 are loaded separately into the naosystem
called PDF nanoparticles to form PDFI and PDFP nanoparticles, and the
combined treatment of PDFI nanoparticles/laser irradiation and PDFP
nanoparticles represents signi
ficant synergistic effects on inhibiting cell
proliferation and inducing cell apoptosis as well as cell cycle arrest at
G2/M phase in MHCC-97H cells [

]. In addition, several researchers
have developed paclitaxel-loaded formulations using a type of self-as-
sembled nanoparticles (P/NPs), followed by conjugated galactosamine
on the prepared nanoparticles (Gal-P/NPs), which appears to have
obviously e
fficacy in the reduction of the size of the tumor [

].
Moreover, functionalized silver nanoparticles (AgNPs) is synthesized by
polyethylenimine as well as paclitaxel. Based on further mechanism
study, the composition-induced HepG2 cell apoptosis gets through
translocation of phosphatidylserine, accumulation of the sub-G1 cells
population, depletion of mitochondrial membrane potential, caspase-3
activation, poly (ADPribose) polymerase cleavage, DNA fragmentation,
and also the signaling pathways involved including AKT, p53 and
MAPK activation [

].
In generally, terpenoids alkaloid plays a role in anti-hepatocarci-
nogenesis by regulating AKT, p53, Caspase-3, MAPK, AFP, Ras and
survivin protein to in
fluence DNA fragmentation, cell apoptosis, pro-
liferation and cell cycle (
Fig. 6
).
2.5. Steroidal alkaloids
Solamargine (a plant derived from steroidal glycoalkaloid) isolated
from Solanum species, shows not only anti-viral, anti-in
flammatory ef-
fects but also antitumor activity in several cancer types including lung
cancer, breast cancer and liver cancer [
116
,

–134
]. Some re-
searchers have shown that the viability and proliferation of HepG2 cells
are repressed by solamargine in a concentration-dependent manner
[
117
]. The expression of EMT relational proteins such as N-ca and vi-
mentin are conformably downregulated, whereas Eca is upregulated in
solamargine treated HCC cells [
117
]. Besides, solamargine can suppress
the migration and invasion of liver cancer cells by down-regulating the
activity of MMP-2 and MMP-9 enzymes [

]. Hence, it is indicated
that solamargine inhibits the migration and invasion of liver cancer
cells by blocking EMT. In addition, other typical mechanisms of sola-
margine against liver cancer might alter DNA content, cell morphology
and gene expression of cells [
119
]. Moreover, after treatment with so-
lamargine, the cells occurred DNA fragmentation, chromatin con-
densation and Sub-G1 cells increase, indicating that solamargine could
induce cell death through irreversible apoptosis [
119
]. Previous ex-
periment has shown that HepG2 and SMMC-7721 cells atrophy and
become rounder and smaller, and cell density is reduced observably
after treatment with solamargine [

]. The results have demonstrated
that solamargine could cause morphological changes of liver cancer
cells and exert dramatically antiproliferative e
ffects on cells [

]. The
increase of Caspase-3 activity in a dose-dependent manner reveals that
solamargine-induced apoptosis is associated with activation of caspase-
3 [
120
]. Interestingly, solamargine-induced apoptosis is related to ar-
resting cell cycle progression, because solamargine arrests HCC cells at
G2/M phase, and mainly facilitates cells from G2/M phase to sub-G1
phase of apoptosis [

]. Besides stimulating caspase-3, solamargine
could also increase of the ratio of Bax to Bcl-2 and decrease of mutation
p53 [

]. Tumor necrosis factor (TNF) serves as a key role in cell
proliferation and cell death as well as in
flammation. It is not only able
to activate the transcription factor NF-
κB, but also could induce ne-
croptosis and apoptosis. But most e
ffects of TNF are mediated through
TNF Receptor 1 (TNFR1) [

]. There are evidences to prove that so-
lamargine might trigger gene expression of human TNFR I in Hep3B
cells [

].
Currently, there are few studies on the use of steroidal alkaloids for
liver cancer. More steroidal alkaloids that are targeted at liver cancer
are still under investigation. Some studies have shown steroidal alka-
loids a
ffect DNA fragmentation, cell apoptosis, proliferation, cell cycle,
necroptosis and metastasis but its pathway involved in anti-
