3.4.2. Đặc điểm sinh học phân tử liên quan đến khả năng kháng rầy
Sau khi đánh giá các chỉ tiêu liên quan đến năng suất, phẩm chất và khả năng kháng rầy nâu của các giống lúa; chúng tôi tiếp tục phân tích gen kháng rầy nâu của các giống lúa nhằm có cơ sở khoa học cho việc giới thiệu giống lúa có năng suất cao, chất lượng tốt và kháng rầy nâu tốt trồng ở Thừa Thiên Huế.
* Xác định gen kháng rầy nâu bph1
Gen kháng rầy nâu bph1 liên kết với chỉ thị STS BpE18-3, nằm trên NST số 12, khoảng cách di truyền là 3.9 cM [67]. Các cây lúa mang gen bph1 có khả năng kháng rầy nâu biotype 1 và 3 nhưng lại nhiễm với biotype 2 [69].
+ Kết quả PCR
Khuếch đại PCR trên DNA tổng số của các giống lúa IRRI 352, BG 367-2, Sài Đường Kiến An và Lốc Nước với cặp mồi BpE18-3. Kết quả hình 3.7 cho thấy tất cả các giống lúa nghiên cứu đều xuất hiện băng DNA khuếch đại với kích thước khoảng 500 bp. Các băng DNA khá rõ chứng tỏ gen kháng rầy bph1 có mặt trong các giống sử dụng trong nghiên cứu này.

Trình tự nucleotide chỉ thị phân tử của giống lúa nghiên cứu được so sánh với trình tự tương ứng trên cơ sở dữ liệu của GenBank (http://www.ncbi.nlm.nih.gov, accession number: AB200354.1). Kết quả so sánh cho thấy chúng tương đồng nhau 95% (Hình 3.8).
