Despite the availability of several therapeutic options, a safer and more effective approach is urgently sought for treatment of cancers. Numerous novel anticancer agents are being discovered from eastern traditional folk medicine and ethno-pharmacology. Besides animals and plants, mushrooms represent a significant source of bioactive compounds for the development of therapeutics by the pharmaceutical industry. It has been shown that mushrooms possess the ability to stimulate mononuclear cell activation, spleen cell proliferation, and cytokine expression; to modulate anti-proliferative effects on many cancers; to regulate cell cycle and induce apoptosis in cancer cell lines.
In the present study, pharmaceutical properties of tropical perennial polypores such as Ganoderma spp., Phellinus spp. were investigated by means of classical and modern molecular analyses using human leukemia cells. It has been proven that heavy metal bioaccumulation of these mushrooms was at nontoxic level for human cell lines and played no role on activities of these mushrooms. At certain concentration, aqueous mushroom extracts showed no cytotoxic effect but anti-proliferative effect due to induction of apoptosis. Besides Ganoderma species, strikingly, our findings indicate that aqueous extract of Phellinus spp. induces apoptosis in HL-60 through the activation of the caspase cascade through both death receptor pathway and mitochondrial pathway. The expression of many apoptotic/anti-apoptotic regulatory proteins has been also ascertained. We also figured out the significant changes in miRNA expression due to our treatment.

Compound K (C-K) là protopanaxadiol ginsenosides Rb1, Rb2 and Rc (các thành phần chính của nhân sâm Hàn Quốc) được chuyển hóa thành hợp chất K (20-O-D-glucopyranosyl-20(S)-protopanaxadiol) bằng các vi khuẩn ruột ở người và chuột có rất nhiều hoạt tính dược học trong in vitro và in vivo. Những nghiên cứu trước đã chứng minh C-K có chức năng phối tử của thụ thể glucocoticoid (GR). Trong nghiên cứu này, chúng tôi đã chứng minh C-K thực hiện vai trò cơ bản đối với quá trình điều hòa zymonsan bắt nguồn từ nấm kích thích đáp ứng viêm của bạch cầu đơn nhân thông qua ức chế tín hiệu thụ thể Dectin-1 và toll like receptor (TLR)2. Những kết quả xử lý bằng C-K đã được so sánh cùng với Dexamethasone (Dex), một phối tử của GR. Quá trình tiết yếu tố hoại thư u (TNF-α), interleukin (IL)-6 và IL12p40, nhưng không phải IL-10, đã bị ức chế ý nghĩa bằng tiền xử lý cùng với C-K. Kết quả tương tự khi thay C-K bằng Dex. Hơn nữa, zymosan kích thích hoạt động của các protein kinase do chất kích thích phân bào hoạt hóa (MAPKs) bao gồm ERK1/2 và p38 đã bị ức chế bằng tiền xử lý bằng C-K hoặc Dex trong tế bào bạch cầu đơn nhân. Quan trọng hơn, khi ức chế hoạt động của Dectin-1 bằng laminarin hoặc TLR2 bằng kháng thể kháng TLR2 đã phá hủy khả năng ức chế của C-K hoặc Dex trong quá trình sản xuất cytokine tiền viêm và hoạt động của MAPK gây ra bởi zymosan. Những kết quả trên đây chỉ ra rằng C-K kích thích hoạt động của thụ thể glucocorticoid điều hòa một số đáp ứng viêm thông qua thụ thể Dectin-1 và TLR2.
Compound K Regulates Zymosan-induced Inflammatory Responses in Human monocytic cells via Dectin-1 receptor and TLR2

Compound K (C-K) (20-O-D-glucopyranosyl-20(S)-protopana-xadiol), which is metabolized from protopanaxadiol ginsenosides Rb1, Rb2 and Rc by intestinal bacteria in humans and rats, has various immunopharmacological activities in vitro and in vivo. Previous studies have demonstrated C-K is afunctional ligand of glucocorticoid receptor (GR). Here, we demonstrate that C-K plays an essential role for regulation of the yeast derived zymosan-induced inflammatory responses by human monocytes through inhibition of Dectin-1- and toll like receptor (TLR)2-dependent signaling. The results induced by C-K were compared with those by dexamethasone (Dex), a well-known GR agonist. The zymosan-mediated secretion of tumor necrosis factor (TNF)-α, interleukin (IL)-6 and IL-12 p40, but not IL-10, were significantly inhibited by pre-treatment with C-K, as similar to those with Dex. In addition, zymosan-induced activation of mitogen-activated protein kinases [MAPKs; extracellular signal-regulated kinase (ERK) 1/2 and p38] was significantly inhibited by pre-treatment with C-K or Dex in human monocytes. Markedly, Blockade of Dectin-1 using laminarin or TLR2 using specific antibodies (anti-TLR2) abrogated the C-K- or Dex-mediated inhibition of zymosan-induced inflammatory cytokine production, MAPK activation. Collectively, these data indicated that GR-induced C-K regulates zymosan-induced inflammatory responses through Dectin-1- and TLR2-dependent manner.
